This variant lies in the MCCC2 gene (transcript NM_022132.5) at the canonical splice donor site of the intron immediately after coding-DNA position 1216, where T is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: MCCC2 c.1216+2T>C is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Several computational tools predict a significant impact on normal splicing: Two predict the variant abolishes a 5 prime splicing donor site; one predict the variant weakens a 5 prime donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 251086 control chromosomes. c.1216+2T>C has been reported in the literature in individuals affected with Methylcrotonyl-CoA Carboxylase Deficiency. These report(s) do not provide unequivocal conclusions about association of the variant with Methylcrotonyl-CoA Carboxylase Deficiency.
