This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 14095, where G is replaced by T; at the protein level this means replaces alanine at residue 4699 with serine — a missense variant. Submitter rationale: Variant summary: TTN c.10363G>T (p.Ala3455Ser) results in a conservative amino acid change located in the I-band region of the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 8.1e-06 in 245772 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.10363G>T in individuals affected with Limb-Girdle Muscular Dystrophy, Type 2J and no experimental evidence demonstrating its impact on protein function have been reported.
