Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 7.2e-05 in 248358 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in TTN causing Dilated Cardiomyopathy (7.2e-05 vs 0.00039), allowing no conclusion about variant significance. c.67822C>T has been observed in an individual suspected of muscular dystrophy who had two other TTN variants also considered to be of uncertain significance and phase of the variants was not determined/unspecified (Radziwonik-Fraczyk_2024). This report does not provide unequivocal conclusions about association of the variant with Dilated Cardiomyopathy or TTN-related muscular dystrophy.
