Uncertain significance for MHC class I deficiency 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001290043.2(TAP2):c.359G>T (p.Ser120Ile), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with TAP2-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces serine with isoleucine at codon 120 of the TAP2 protein (p.Ser120Ile). The serine residue is moderately conserved and there is a large physicochemical difference between serine and isoleucine. This variant is not present in population databases (ExAC no frequency).
