NM_000368.5(TSC1):c.3434C>T (p.Pro1145Leu) was classified as Uncertain Significance for Tuberous sclerosis syndrome by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015: This missense variant replaces proline with leucine at codon 1145 of the TSC1 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). A transfection-based functional study measuring TSC1 abundance, and downstream p70 S6 kinase (S6K) phosphorylation indicated that this variant does not disrupt TSC1 protein function (PMID: 22903760) . This variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has been identified in 2/251390 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
