NM_001371279.1(REEP1):c.353T>G (p.Leu118Arg) was classified as Likely pathogenic for Hereditary spastic paraplegia 31 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 118 of the REEP1 protein (p.Leu118Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with distal hereditary motor neuropathy (internal data). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 534220). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.
