This variant lies in the MYH11 gene (transcript NM_002474.3) at coding-DNA position 3389, where A is replaced by G; at the protein level this means replaces glutamic acid at residue 1130 with glycine — a missense variant. Submitter rationale: This sequence change replaces glutamic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 1137 of the MYH11 protein (p.Glu1137Gly). This variant is present in population databases (no rsID available, gnomAD 0.004%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with MYH11-related conditions. ClinVar contains an entry for this variant (Variation ID: 534146).
