Uncertain significance for Myoclonic epilepsy, juvenile, susceptibility to, 1; Absence seizure — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_018100.4(EFHC1):c.1147C>T (p.Pro383Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with serine, which is neutral and polar, at codon 383 of the EFHC1 protein (p.Pro383Ser). This variant is present in population databases (rs546262142, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with EFHC1-related conditions. This missense change has been observed in at least one individual who was not affected with EFHC1-related conditions (Invitae). ClinVar contains an entry for this variant (Variation ID: 534110). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
