Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000142.5(FGFR3):c.436_445+2del, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the FGFR3 gene (transcript NM_000142.5) at coding-DNA position 436 through the canonical splice donor site of the intron immediately after coding-DNA position 445, deleting this region. Submitter rationale: Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site, but this prediction has not been confirmed by published transcriptional studies. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. The current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in FGFR3 cause disease. This variant has not been reported in the literature in individuals with FGFR3-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change affects a donor splice site in intron 4 of the FGFR3 gene. It is expected to disrupt RNA splicing and/or create a premature translational stop signal, and likely results in an absent or disrupted protein product.
