Uncertain Significance for Hypertrophic cardiomyopathy — the classification assigned by All of Us Research Program, National Institutes of Health to NM_016203.4(PRKAG2):c.541A>C (p.Lys181Gln), citing ACMG Guidelines, 2015. This variant lies in the PRKAG2 gene (transcript NM_016203.4) at coding-DNA position 541, where A is replaced by C; at the protein level this means replaces lysine at residue 181 with glutamine — a missense variant. Submitter rationale: Variant of Uncertain Significance due to insufficient evidence: This missense variant replaces lysine with glutamine at codon 181 of the PRKAG2 protein. Computational prediction tools and conservation analyses suggest that this variant may not impact the protein function. Computational splicing tools suggest that this variant may not impact RNA splicing. To our knowledge, functional assays have not been performed for this variant nor has this variant been reported in individuals affected with cardiovascular disorders in the literature. This variant has been identified in 3/277234 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Available evidence is insufficient to determine the pathogenicity of this variant conclusively.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
