This variant lies in the SDHD gene (transcript NM_003002.4) at coding-DNA position 82, where C is replaced by G; at the protein level this means replaces proline at residue 28 with alanine — a missense variant. Submitter rationale: This sequence change replaces proline with alanine at codon 28 of the SDHD protein (p.Pro28Ala). The proline residue is moderately conserved and there is a small physicochemical difference between proline and alanine. This variant is not present in population databases (ExAC no frequency). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
