As the precise location of this duplication is unknown, it may be in tandem or it may be located elsewhere in the genome. Duplication of PLP1 is clearly defined as a Pelizaeus-Merzbacher disease causative allele and is the most common cause of disease in affected individuals (PMID: 9633722, 10417279, 8659540, 10319885, 23347225). For these reasons, this variant has been classified as Pathogenic.
