Uncertain significance for Familial adenomatous polyposis 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001048174.2(MUTYH):c.253T>C (p.Trp85Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tryptophan, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 113 of the MUTYH protein (p.Trp113Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with colorectal cancer and/or polyps who also carried another pathogenic variant in MUTYH (PMID: 28644590). ClinVar contains an entry for this variant (Variation ID: 533303). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive.
