Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004655.4(AXIN2):c.1642G>A (p.Glu548Lys), citing Ambry Variant Classification Scheme 2023: The p.E548K variant (also known as c.1642G>A), located in coding exon 5 of the AXIN2 gene, results from a G to A substitution at nucleotide position 1642. The glutamic acid at codon 548 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
