Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000492.4(CFTR):c.1597T>C (p.Phe533Leu), citing LabCorp Variant Classification Summary - May 2015: Variant summary: CFTR c.1597T>C (p.Phe533Leu) results in a non-conservative amino acid change located in the ABC transporter-like, ATP-binding domain (IPR003439) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 250892 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1597T>C has been reported in the literature in at least one individual affected with Cystic Fibrosis, however the second CFTR variant in this individual remains unknown (e.g., Kambouris_2000). This report therefore does not provide unequivocal conclusions about association of the variant with Cystic Fibrosis. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 10834512, 22326559). No submitters have reported clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
