NM_000388.4(CASR):c.1087G>A (p.Gly363Ser) was classified as Uncertain significance for Nephrolithiasis/nephrocalcinosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G363S variant (also known as c.1087G>A), located in coding exon 3 of the CASR gene, results from a G to A substitution at nucleotide position 1087. The glycine at codon 363 is replaced by serine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
