Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004360.5(CDH1):c.221G>T (p.Arg74Leu), citing Ambry Variant Classification Scheme 2023: The p.R74L variant (also known as c.221G>T), located in coding exon 3 of the CDH1 gene, results from a G to T substitution at nucleotide position 221. The arginine at codon 74 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
