Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004360.5(CDH1):c.2593T>G (p.Trp865Gly), citing Ambry Variant Classification Scheme 2023: The p.W865G variant (also known as c.2593T>G), located in coding exon 16 of the CDH1 gene, results from a T to G substitution at nucleotide position 2593. The tryptophan at codon 865 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
