This variant lies in the RYR2 gene (transcript NM_001035.3) at coding-DNA position 7225, where A is replaced by T; at the protein level this means replaces isoleucine at residue 2409 with phenylalanine — a missense variant. Submitter rationale: This variant is not present in population databases (ExAC no frequency). This sequence change replaces isoleucine with phenylalanine at codon 2409 of the RYR2 protein (p.Ile2409Phe). The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and phenylalanine. This variant has not been reported in the literature in individuals with RYR2-related disease.
