NM_058195.4(CDKN2A):c.149T>G (p.Leu50Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L50R variant (also known as c.149T>G), located in coding exon 1 of the CDKN2A (p14ARF) gene, results from a T to G substitution at nucleotide position 149. The leucine at codon 50 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
