Pathogenic for Diamond-Blackfan anemia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000001.11:g.(?_92832109)_(92833666_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in RPL5 are known to be pathogenic (PMID: 19773262, 19061985). This variant has not been reported in the literature in individuals with RPL5-related disease. This variant is a gross deletion of the genomic region encompassing exons 1-3 of the RPL5 gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 3 of the RPL5 gene. This is expected to result in an absent or disrupted protein product.
