Uncertain significance for Congenital myotonia, autosomal recessive form; Congenital myotonia, autosomal dominant form — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000083.3(CLCN1):c.2239C>T (p.Pro747Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline with serine at codon 747 of the CLCN1 protein (p.Pro747Ser). The proline residue is highly conserved and there is a moderate physicochemical difference between proline and serine. This variant is present in population databases (rs373850192, ExAC 0.002%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with CLCN1-related disease.
