Pathogenic for Hereditary breast ovarian cancer syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.11:g.(?_43074325)_(43115785_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exons 3-14 of the BRCA1 gene. This creates a premature translational stop signal and is expected to result in an absent or disrupted protein product. A similar deletion of exons 3-14 (described as deletion of exons 3-15) has been reported in the literature in an affected individual from a hereditary breast and ovarian cancer family (PMID: 12960223). Exon 14 of BRCA1 is also known as exon 15 in the literature. Loss-of-function variants in BRCA1 are known to be pathogenic (PMID: 20104584). For these reasons, this variant has been classified as Pathogenic.
