This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 3194, where A is replaced by T; at the protein level this means replaces aspartic acid at residue 1065 with valine — a missense variant. Submitter rationale: This sequence change replaces aspartic acid with valine at codon 1065 of the BRCA1 protein (p.Asp1065Val). The aspartic acid residue is weakly conserved and there is a large physicochemical difference between aspartic acid and valine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with BRCA1-related disease.
