Uncertain Significance for Breast-ovarian cancer, familial, susceptibility to, 1 — the classification assigned by All of Us Research Program, National Institutes of Health to NM_007294.4(BRCA1):c.5347A>G (p.Met1783Val), citing ACMG Guidelines, 2015: This missense variant replaces methionine with valine at codon 1783 of the BRCA1 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function. A functional study has reported that this variant does not impact BRCA1 function in a haploid cell proliferation assay (PMID: 30209399). This variant has been reported in two individuals affected with breast or ovarian cancer (PMID: 28664449, 34235180). This variant has been identified in 1/251446 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
