This variant lies in the NBN gene (transcript NM_002485.5) at coding-DNA position 1327, where A is replaced by G; at the protein level this means replaces lysine at residue 443 with glutamic acid — a missense variant. Submitter rationale: This sequence change replaces lysine with glutamic acid at codon 443 of the NBN protein (p.Lys443Glu). The lysine residue is weakly conserved and there is a small physicochemical difference between lysine and glutamic acid. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with NBN-related disease.
