Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000465.4(BARD1):c.1909A>G (p.Lys637Glu), citing Ambry Variant Classification Scheme 2023: The p.K637E variant (also known as c.1909A>G), located in coding exon 10 of the BARD1 gene, results from an A to G substitution at nucleotide position 1909. The lysine at codon 637 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
