NM_024675.4(PALB2):c.157G>A (p.Glu53Lys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Sema4, Sema4, citing Sema4 Curation Guidelines: To the best of our knowledge, the PALB2 c.157G>A (p.E53K) variant has not been reported in individuals with PALB2-related disease. It has been reported in a large case-control study of breast cancer in 1/60466 cases and 0/53461 controls (PMID: 33471991). It was observed in 1/34592 chromosomes of the Latino subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has been reported in ClinVar (Variation ID 530120). In silico tools suggest the impact of the variant on protein function is benign, though these predictions have not been confirmed by functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
