NM_024675.4(PALB2):c.1908G>C (p.Glu636Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E636D variant (also known as c.1908G>C), located in coding exon 5 of the PALB2 gene, results from a G to C substitution at nucleotide position 1908. The glutamic acid at codon 636 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
