This variant lies in the BARD1 gene (transcript NM_000465.4) at coding-DNA position 1918, where C is replaced by A; at the protein level this means replaces leucine at residue 640 with isoleucine — a missense variant. Submitter rationale: The p.L640I variant (also known as c.1918C>A), located in coding exon 10 of the BARD1 gene, results from a C to A substitution at nucleotide position 1918. The leucine at codon 640 is replaced by isoleucine, an amino acid with highly similar properties. This variant was seen in 1/1058 patients with colorectal cancer (Yurgelun MB et al. J. Clin. Oncol., 2017 Apr;35:1086-1095). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive.
