This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 994, duplicating one base; at the protein level this means shifts the reading frame starting at isoleucine residue 332, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.994dupA pathogenic mutation, located in coding exon 9 of the BRCA2 gene, results from a duplication of A at nucleotide position 994, causing a translational frameshift with a predicted alternate stop codon (p.I332Nfs*4). This alteration was identified in a multiple studies of BRCA1/2 mutation positive families (Lecarpentier J et al. Breast Cancer Res, 2012 Jul;14:R99; Rebbeck TR et al. Hum Mutat, 2018 05;39:593-620). In addition to the clinical data presented in the literature, his alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay.
