Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000093.5(COL5A1):c.2744C>T (p.Thr915Met), citing Ambry Variant Classification Scheme 2023: The p.T915M variant (also known as c.2744C>T), located in coding exon 33 of the COL5A1 gene, results from a C to T substitution at nucleotide position 2744. The threonine at codon 915 is replaced by methionine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
