Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_004260.4(RECQL4):c.903A>T (p.Glu301Asp), citing Sema4 Curation Guidelines. This variant lies in the RECQL4 gene (transcript NM_004260.4) at coding-DNA position 903, where A is replaced by T; at the protein level this means replaces glutamic acid at residue 301 with aspartic acid — a missense variant. Submitter rationale: The RECQL4 c.903A>T (p.E301D) variant has not been reported in the literature to our knowledge. It was observed in 1/111992 chromosomes of the Non-Finnish European subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org). The variant has been reported in ClinVar (Variation ID 529003). In silico tools suggest the impact of the variant on protein function is benign, though these predictions have not been confirmed by functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
