Pathogenic for Melnick-Fraser syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000503.6(EYA1):c.532del (p.Ala178fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in EYA1 are known to be pathogenic (PMID: 18220287). This variant has not been reported in the literature in individuals with EYA1-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Ala178Hisfs*63) in the EYA1 gene. It is expected to result in an absent or disrupted protein product.
