This variant lies in the SDHB gene (transcript NM_003000.3) at coding-DNA position 819, where T is replaced by A; at the protein level this means converts the codon for tyrosine at residue 273 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with SDHB-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change results in a premature translational stop signal in the SDHB gene (p.Tyr273*).
