This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 9507, deleting one base; at the protein level this means shifts the reading frame starting at isoleucine residue 3169, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: Variant summary: BRCA2 c.9507delT (p.Ile3169MetfsX48) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. The variant was absent in 251302 control chromosomes. c.9507delT has been reported in the literature in individuals affected with Hereditary Breast And Ovarian Cancer Syndrome (e.g., Fries_2002, Rebbeck_2018). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 11873550, 29446198). Three ClinVar submitters (evaluation after 2014) have cited the variant, and all laboratories classified the variant as pathogenic.
