NM_001999.4(FBN2):c.1901A>G (p.Asn634Ser) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N634S variant (also known as c.1901A>G), located in coding exon 14 of the FBN2 gene, results from an A to G substitution at nucleotide position 1901. The asparagine at codon 634 is replaced by serine, an amino acid with highly similar properties, and is located in the cbEGF-like #06 domain. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
