NM_001999.4(FBN2):c.4959A>C (p.Glu1653Asp) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1653D variant (also known as c.4959A>C), located in coding exon 39 of the FBN2 gene, results from an A to C substitution at nucleotide position 4959. The glutamic acid at codon 1653 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
