Pathogenic for Spastic paraplegia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000023.10:g.(?_153132791)_(153141311_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exons 1-16 of the L1CAM gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 16 of the L1CAM gene. This is expected to result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in L1CAM are known to be pathogenic (PMID: 19846429). This variant has not been reported in the literature in individuals with L1CAM-related disease.
