Uncertain significance for Neuronal ceroid lipofuscinosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_025219.3(DNAJC5):c.238G>A (p.Gly80Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 80 of the DNAJC5 protein (p.Gly80Ser). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with DNAJC5-related conditions. This missense change has been observed in at least one individual who was not affected with DNAJC5-related conditions (internal data). ClinVar contains an entry for this variant (Variation ID: 527738). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
