While the exact position of the duplicated exons cannot be determined from this data, sub-genic duplications are generally in tandem (PMID: 25640679) and may result in an absent or disrupted protein product. This variant has not been reported in the literature in individuals with NF1-related disease. However, itt has been found to segregate with NF1-related disease in a single family (Invitae). Experimental studies have not been reported for this copy number gain and it is currently uncertain if it results in a disrupted protein. Loss-of-function variants in NF1 are known to be pathogenic (PMID: 10712197, 23913538). For these reasons, this variant has been classified as Pathogenic.
