Loss-of-function variants in NF1 are known to be pathogenic (PMID: 10712197, 23913538). This variant has not been reported in the literature in individuals with NF1-related disease. A different nucleotide change that causes the same premature translational stop signal (c.7365T>G) has been reported in an individual affected with neurofibromatosis type 1, in the Leiden Open-source Variation Database (PMID: 21520333). ClinVar contains an entry for this variant (Variation ID: 527542). This variant is not present in population databases (ExAC no frequency).
