This variant lies in the NF1 gene (transcript NM_001042492.3) at coding-DNA position 311 through coding-DNA position 312, deleting 2 bases. Submitter rationale: This variant is not present in population databases (ExAC no frequency). For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in NF1 are known to be pathogenic (PMID: 10712197, 23913538). Two different variants (c.311T>A, c.311T>G) giving rise to the same protein effect observed here (p.Leu104*) have been reported in individuals affected with neurofibromatosis type 1 (PMID: 15060124, 16944272, 27838393). This variant has not been reported in the literature in individuals with NF1-related disease. This sequence change creates a premature translational stop signal (p.Leu104*) in the NF1 gene. It is expected to result in an absent or disrupted protein product.
