Pathogenic for Neurofibromatosis, type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001042492.3(NF1):c.6402T>A (p.Cys2134Ter), citing Invitae Variant Classification Sherloc (09022015): This variant has been reported in an individual affected with neurofibromatosis type 1 (PMID: 7655472). ClinVar contains an entry for this variant (Variation ID: 527417). Loss-of-function variants in NF1 are known to be pathogenic (PMID: 10712197, 23913538). For these reasons, this variant has been classified as Pathogenic. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Cys2113*) in the NF1 gene. It is expected to result in an absent or disrupted protein product.
