Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.779A>G (p.His260Arg), citing Ambry Variant Classification Scheme 2023: The p.H260R variant (also known as c.779A>G), located in coding exon 6 of the RAD50 gene, results from an A to G substitution at nucleotide position 779. The histidine at codon 260 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
