This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 3408, where C is replaced by A; at the protein level this means converts the codon for tyrosine at residue 1136 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change creates a premature translational stop signal (p.Tyr1136*) in the FBN1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in FBN1 are known to be pathogenic (PMID: 17657824, 19293843). This variant is not present in population databases (gnomAD no frequency). A different variant (c.3408C>G) giving rise to the same protein effect has been determined to be pathogenic (PMID: 18435798).
