Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_004364.5(CEBPA):c.432G>T (p.Glu144Asp), citing Ambry Variant Classification Scheme 2023: The p.E144D variant (also known as c.432G>T), located in coding exon 1 of the CEBPA gene, results from a G to T substitution at nucleotide position 432. The glutamic acid at codon 144 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
