Pathogenic for Hereditary nonpolyposis colorectal neoplasms — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000002.12:g.(?_47795888)_(47803691_?)del, citing Invitae Variant Classification Sherloc (09022015): Loss-of-function variants in MSH6 are known to be pathogenic (PMID: 18269114, 24362816). For these reasons, this variant has been classified as Pathogenic. Exon 3-5 deletions have not been reported in the literature in individuals with MSH6-related disease. This variant is an out-of-frame deletion of the genomic region encompassing exons 3-5 of the MSH6 gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product.
