This variant lies in the MSH6 gene (transcript NM_000179.3) at coding-DNA position 3160, where A is replaced by G; at the protein level this means replaces isoleucine at residue 1054 with valine — a missense variant. Submitter rationale: The p.I1054V variant (also known as c.3160A>G), located in coding exon 4 of the MSH6 gene, results from an A to G substitution at nucleotide position 3160. The isoleucine at codon 1054 is replaced by valine, an amino acid with highly similar properties. This alteration was detected on a 25-gene panel test in a woman who was diagnosed with breast cancer before age 50 (Tung N et al. Cancer, 2015 Jan;121:25-33). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
