Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000179.3(MSH6):c.3305C>G (p.Thr1102Ser), citing Ambry Variant Classification Scheme 2023: The p.T1102S variant (also known as c.3305C>G), located in coding exon 5 of the MSH6 gene, results from a C to G substitution at nucleotide position 3305. The threonine at codon 1102 is replaced by serine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
